![]() Approximately 80% of all captured cells are proximal and collecting tubule origin, which highlights the role of the renal tubular system in health and disease (Park et al, 2018). By using this revolutionary technique, the first scRNA-seq analysis in kidneys of C57BL/6 mouse is performed and has generated a comprehensive cell atlas and identified 19 distinct cell types from normal murine kidneys. Single-cell RNA sequencing (scRNA-seq) analysis can redefine cell types in tissues and identify the cell-type specific changes of gene expression (Tang et al, 2009 Macosko et al, 2015 Zheng et al, 2017). This urgent event requires us to reveal more accurate molecular mechanism of DKD to develop more efficacious treatment in this disease. Such approaches might show the delayed onset of renal failure, but many patients still progress to end-stage kidney failure (ESKF, also known as end-stage renal disease) despite optimal therapy with the current state of medical care (Thomas et al, 2015 Doshi & Friedman, 2017 Anders et al, 2018). Currently, intensive management of patients with DKD includes controlling blood glucose levels and blood pressure as well as blockade of the renin–angiotensin–aldosterone system (RAAS). T2D is the leading cause of impaired kidney function, albuminuria, and renal replacement therapy (dialysis or transplantation) worldwide, thus placing a large burden on health-care systems (Thomas et al, 2016, 2015 Doshi & Friedman, 2017). Diabetic kidney disease (DKD, previously named as diabetic nephropathy) is a microvascular complication and progresses gradually over many years in approximately 50% of patients who are T2D (Thomas et al, 2016). Of DM, type 2 diabetes (T2D) is the most common form (more than 90%) and characterized by insulin resistance and abnormalities in insulin production. According to the latest report from the International Diabetes Federation (IDF), approximately 463 million adults (20–79 years) were living with diabetes in 2019, while this will rise to 700 million by 2045 ( ). ![]() Diabetes mellitus (DM) is approaching epidemic proportions globally.

The SDD-3000 is no one-trick-pony though, as it has plenty of useful controls under its (digital) hood: the input can be pushed to create a driven sound that ranges from warm to crunchy, the regeneration section features low and high filters to tweak the timbre of your repeats and the modulation section can be utilized to give your repeats a subtle warble or a wild, alien-like effect. Whereas the aforementioned delay plugins allow you to access many different delay types, the Korg SDD-3000 from Universal Audio is a faithful emulation of a hardware unit that was released in 1982 and popularized by The Edge (guitarist for U2). Also, don’t sleep on FreqEcho, which is a simple but free delay plugin available from Valhalla DSP. When you switch modes or hover over any other parameter of Valhalla Delay, the plugin offers you a handy block of text towards the bottom of the GUI which explains the parameter. Valhalla Delay is actually an excellent learning tool if you want to know more about delay and how specific controls affect the overall sound. It also has controls over modulation, drive, EQ and more for fine-tuning the sounds of your delay. Valhalla Delay features 10 different delay modes including tape, HiFi, digital and more. They sound fantastic, the user interfaces are clean looking and they are designed in such a way to encourage a sense of play. I’m a huge fan of all the plugins from Valhalla DSP. I’ll map parameters (usually the feedback, mix and filters) to my MIDI controller, and “play” the delay like it’s a musical instrument. This is something I’ll do with all types of plugins to give my processors a greater degree of musicality, and EchoBoy is no exception. Sometimes I’ll even use EchoBoy only as a “color unit,” as the saturation and filters sound fantastic when combined with the unique timbers of the 30 styles. EchoBoy excels at creating delay on virtually any type of signal including vocals, synths, guitars and beyond. Combine these styles with the onboard high and low-cut filters, saturation and input & output controls, and you can sculpt the character of your repeats to be crystal clear, warbly, warm, dull, brash, etc. At the heart of EchoBoy are 30 distinct styles of delay which are based on classic gear, making this plugin a veritable history lesson on delay in addition to a fantastic music production utility. It’s got everything you need in a delay and more. While I utilize each and every delay plugin listed in this roundup, I could likely make do using only EchoBoy. Here are my favorite plugins to use while mixing. Today, we benefit from software companies having created plugin emulations of vintage hardware delay units, while also creating digital tools that are far more complex and controllable than any mechanical delay hardware could ever be. Rack mountable studio gear came next, with processors such as the Korg SDD-3000 and Lexicon Prime Time Delay defining the sound of the 1980’s. ![]() Eventually, players and engineers began using self-contained delay units such as the Binson Echorec, the Echoplex and the Roland Space Echo. It would take a certain amount of time for that signal to be heard, hence the term delay. A signal being captured on one tape recorder was sent to another recorder, which was subsequently sent back to the first recorder. ![]() The history of incorporating delay in music production is an interesting one, and can be traced back to the practice of using multiple magnetic tape recorders to create an echo effect. It can be utilized creatively to create a droning ambient tail, you can use a single repeat set to a 40-100 millisecond delay to pay homage to early rock and roll or you can set up a ping-pong “ throw delay” to make your pop mix sound more produced and “expensive.” These are just some of the many ways you can incorporate delay into your mixes. When used subtly it can establish a sense of space in which tracks can sit, similar to reverb. When producing and mixing music, delay is such a versatile effect. ![]() ![]() The result is a more complicated process in which the seller needs to get permission to start the process and doesn’t have control of how or when it ends because the lender decides which offer to accept.įollow these two steps if you think you might need to do a short sale. While most home sales occur without the knowledge of the lender until the sales process is in its late stages, the lender is involved from the beginning of a short sale. The most significant difference between a short sale and a typical home sale is the lender’s central role in the short sale. If you’re part of that small minority, though, a short sale could prove helpful. Short sales aren’t nearly as common today, as just 3% of all mortgaged properties, or 1.6 million homes, had negative equity in the third quarter of 2020, according to CoreLogic, a property analytics firm. Homeowners who could no longer afford payments often were forced into short sales. Short sales were more prevalent during the Great Recession when home values crashed in many parts of the country, leaving struggling homeowners with negative equity. A short sale is often a shorter process than foreclosure, but there’s no guarantee your lender will agree to this option.Įven though a short sale is similar to a typical home sale in certain ways–you would likely rely on a real estate agent to list the property, and it could be sold to a conventional buyer who finds it online–it takes considerably more paperwork and your lender will be heavily involved. A short sale occurs when you sell your home for less than you owe on the mortgage, and your lender forgives the outstanding balance instead of pursuing a foreclosure case. ![]() In PD, since L/R audio channels are never separated or exposed to the editor, you can't normalize a single audio source as I described prior. If you select more than one clip in the "Mix" area as I showed in the AD pic I attached, you are normalizing between clips. Not really something one typically wants to do as that's what creates part of the stereo atmosphere. Just remember, when you do this in AD, if you normalize a single audio clip in just the "Edit" mode by selecting both channels, you are essentially balancing the left and right channels of that clip if stereo audio. As I used to teach in the military, where there's a will, there's a way. I'm not a quitter and can't accept "it can't be done". I'm determined to get this done come Hell or high water. Once I get that done, I'm going to play with them and PD and I'm also gonna play with the audio clips on AD. I have some new video clips but they have foreign titles so I'll have to do some translating which is a bummer. I deleted all my original video clips and was working with just audio clips. As usual more absolutely irreplaceable info. Both clips are normalized to each other so I don't have to constantly adjust the volume knob though. Here things are normalized to the lower level so I would potentially have to boost the gain of the entire track or during playback I'd have to turn up the volume vs typical. You can see similar effects here, Track 1 original, Track 2 normalized. The concept in AD is about the same as shown in attached pic, just not as easy working with multiple clips as you have to bring them in. I'll be back and give an update on how good I am at following directions. ![]() Actually, I'm about to delete that clip along with the others that are too low. I've been using Audio Director after everything else failed. Quote The PD "Normalize" works on the selected clips to adjust gain between clips, it's not applicable to a single clip as there is nothing to normalize, the gain is what it is and could be lowered or raised but not normalized.Īh so. All clips are normalized to each other so you don't want to tweak a clip independently, hence the horizontal slider to adjust the whole track gain. If you find that this normalized volume is too low overall on track 1, use the horizontal slider to boost the gain of the entire track. normalized), no longer normal, soft, boost as was previous. Now all 3 of these clips will have the same level when played (i.e. When one selects all 3 clips and uses the "Normalize" feature you can see the gain of clip 2 was increased significantly, clip 3 was actually lowered a little, these are shown in track 1. So if I'd just produce track 2 one would have clip 1 that maybe sounds normal, clip 2, very soft, clip 3 a little bit of a volume boost. Notice the very low audio level of the middle clip. In the attached pic, track 2 is the original clips. The PD "Normalize" works on the selected clips to adjust gain between clips, it's not applicable to a single clip as there is nothing to normalize, the gain is what it is and could be lowered or raised but not normalized. I'm pretty sure I'm not understanding your link. Then, because it's a playlist, I then do an album gain. I then do a track analysis and it adjusts the tracks to that db. In my other efforts I've run the 24 clips through MP3Gain, target db is 89.0. Go to the file where the clip is, volume still low. Produce it and when I click Save it asks if I want to overwrite existing file. It's already normalized so, I go to the horizontal line below the clip and increase it to 4.78db. I imported the clip that has been bugging me. Adjust and check until you get them close. If so, determining how much to increase is a "crap shoot". Once I "normalize" all the clips then I can check to see if any are still lower in playback vollume and if so, open those links and with the horizontal line adjust the volume up. Quote Maybe this will help you about normalize in PD. If you need amplifying info, let me know. So, my question - is it possible that an audio clip cannot be modified? However, when I click on Save or Produce, then go to the saved/produced link, still original volume. In all above instances, when I clicked on the Play in each instance, the clip was loud. Same result.ĭesperate, I installed the free version of Cyberlink Audio Director. So, I used AVS Audio Editor to normalize. Because all 24 have different playback volumes, I used MP3 GainGUI normalize them. I've used Pazera to extract the audio on all 24. I've downloaded a playlist with 24 music videos. I'm using an HP desktop, Win 10, i7 360Hz, 16mb mem, 1tb hdd, Chrome. I then tried to get the volume of that track to match the volume of 23 other audio tracks. ![]() I downloaded a music video from You Tube, used Pazera to extract the audio. Is it possible for an audio track to not be normalized? ![]() You see your uncle as a person and not just as a plot device due to this, and it works to motivate you to get past the tough challenges that await. I think the early home-searching section plays a large part in that since it does go on for a while and absorbs you into the world. While the story isn’t the most captivating you’ll ever see in a game, it is among the best ones yet in a platformer and does make you care about the characters fairly quickly. You get a sense of panic at times worrying about Fred, and worry about the frog people because they all seem so naive and innocent. There’s a lot of dialogue here, and while there’s definitely some damage done by it seeming like the game’s creators don’t natively speak the language, the story is still emotional. The narrative-heavy structure was a surprise, and a pleasant one. Being able to use the mouse to aim and the left stick to move is nice though, and was a fine combination that allowed me to tackle some of the tougher challenges. There’s also a keyboard and mouse setup, and while the mouse is far better for aiming, I found the dual stick setup to be more user-friendly - but I’m also a casual PC gamer and stick to gamepad controls whenever possible. It’s a comfortable setup that ensures you’ll always hit the button you want to when you need it. Holding down RB charges your jump, while LB runs and RT uses the grappling hook and A jumps. The control scheme is logical and easy to remember at all times during the game. Timing is also crucial, as you’ll be in mid-air for a lot of the game and won’t have the pseudo net of a platform beneath you to cushion your fall. You’ll also need to use your noodle for things like the correct angle and height for a jump before using the grappling hook. Sure, there are some brain teasers here - but they’re limited to figuring out just the right amount of momentum to have for a particular leap of faith. ![]() When you first lay eyes on it, you might think there’s a bit of Portal here with all the ambient lighting but there’s not. This is also more of a platformer than those games ever were and it sticks to that term fairly literally for much of the adventure. It’s a bit like Metroid or Bionic Commando, but the first-person perspective mixes things up a bit. ![]() ![]() All you know is your uncle is missing and this strange race of frog people have turned up at around the same time, so you wind up wondering if they did anything to him despite them seeming to be honest on the surface.ĪSAMU’s power suit increases the strength and agility of anyone who wears it, and it comes with a nifty energy beam grappling hook. Everyone is concerned about them and the village’s elder will even give you a giant crystal to upgrade your suit. They haven’t seen him in a while, but want to help you out if they can. You meet with Maddie – a young frog girl who lives in a village and knows your uncle. The challenge starts off quickly thanks to an obstacle course that doubles as a tutorial, which transitions nicely into continuing the narrative. You’ve got a fairly broad skillset to start the game off with as a basic jump, a power jump, and the grappling hook are available right away. Where that game focused on parkour-platforming though, this one focuses on precision platforming that tests your mettle quickly. It’s something we haven’t seen much of outside of Mirror’s Edge, and while it isn’t as stylized as that, it does succeed in offering a similarly-thrilling experience. This game is a rarity - a first-person platformer. This is where you learn the game’s key mechanics and first get hooked on the game’s core design. This suit allows you to swing around and wouldn’t you know it - that comes in handy when you’re transported into a mysterious world that doubles giant obstacle course. You did, however, find his observatory alongside a custom-fitted power suit he created just for you. You began by examining his home, but have no luck finding him. This adventure begins in a rather absorbing way - with your character talking to his daughter about an adventure in his younger days searching for his uncle Fred. This one's easy: most of the same features you’d consider when buying any grooming device-cutting power, ease-of-use, battery life-are key here too, though ergonomics factor heavily into the equation (and safety-enhancing features like extra-fine blades and wet-dry capabilities for a quick clean should be a prerequisite, too). What we're trying to say is: yes, you really do need one, no matter what type of awkward, halfhearted assurances you’ve heard in the past. (Just ask de Bergerac! ) So don't rely on the perceptive ability of your friends/family/closest Zoom confidants-seize the initiative yourself. Unlike, say, a brand new set of whiskers or an unruly pair of sideburns, they're not all that noticeable until, suddenly, they really are soliciting an honest opinion about the state of your schnoz is a surefire way to catalyze an uncomfortable bout of hem-hawing from everyone within a few-mile radius. They're also especially important to pay attention to yourself because, well, they're extremely awkward for anyone else to bring up. We're all friends here, so we're going to level with you: even if your default look is “rakishly unkempt," errant nostril hairs are never a great look. Sticking just anything else in there is ill-advised at best, and downright dangerous at worse. (Never mind trying to wedge some scissors up there.) To properly tame any errant nose and ear hairs, you're going to want a machine specifically designed to get up in the contours of your nostrils. The shaver you use to tame your facial hair isn’t going to cut it-nor will the clippers you rely on for at-home touch-ups. Which means it's worth picking up a dedicated gadget to do the job. A good nose and ear hair trimmer will cut your hairs down to one standard length, keeping them well out of view while still in prime functioning and filtering condition. You don't want to go scorched earth and simply pluck them into oblivion. Namely, filtering out all the dirt and dust in the air you inhale every day, so you're not just sucking grime into your lungs. But there's a really good reason to avoid plucking your way to a well-groomed schnoz: your nostril hairs serve a distinct purpose. It might seem like even the best nose hair trimmer is a needless indulgence for anyone who already owns tweezers. |
